Well if you've read my thread on CS6 you already know how I feel. However, for the moment I'm still on CS5.5 but do plan on getting CS6 (along with a bundle of new software) by the end of next month.
I downloaded the trial a while back and found enough new stuff that I was impressed. Once you've used the new features and applied all the new graphic accelerated effects, you won't want to go back.
Still, I'd be remiss to imply that there's a dramatic difference in everything (Photoshop notwithstanding) its just that with CS6 most everything's a whole lot easier--and therefore less time consuming--to do.
I wouldn't change during an ongoing project.

One of the first experiments I tried. When I opened some of my CS5.5 project in CS6 I didn't experience anything anomalies. All effects, transition, edits, metadata etc.were transferred over and worked without a hitch. However, that was just with my work and on a limited basis. There are hundreds of effects and trying that with every one of them would take months. If you have to do it, do it early.
You might also be interested to know that when I did the same thing transitioning from CS4 to CS5.5, I did discover one error but that was in AA. Things get a lot more complicated when you're compositing and layering one feature on top of another. One feature that was available in CS4 had been discontinued by CS5. No doubt that was because it was going from a 32bit program to a 64bit one. A much bigger leap.
